Multiple forms of deoxythymidine kinase in various tissues.

چکیده
A simple method to detect the existence of multiple forms of deoxythymidine was established. With this method, the pattern of the enzyme was examined in various normal and tumor tissues. The existence of multiple forms of the enzyme was demonstrated in tumor tissues such as Yoshida sarcoma, Ehrlich ascites, Hepatoma AH 130, Sarcoma 180, and Morris Hepatomas 7793 and 7794A, but not in normal rat tissues such as regenerating and embryonic livers and marrow cells.

Distribution and regulation of deoxythymidine kinase activity in differentiating cells of mammalian intestines.
SUMMARY The distribution of deoxythymidine kinase activity has been reexamined in rat and human intestinal mucosa separated by a planing technique into cells in different stages of differentiation. When epithelial cells leave the crypts, a decrease in kinase activity was observed in both resuspended pellet and supernatant fractions. Commercial preparations of phospholipase C added to the assay ...
متن کاملHerpesviral deoxythymidine kinases contain a site analogous to the phosphoryl-binding arginine-rich region of porcine adenylate kinase; comparison of secondary structure predictions and conservation.
Twelve herpesviral deoxythymidine kinases were examined for regions of sequence similarity by multiple alignment. Six highly conserved sites were observed. Site 1 corresponded to a glycine-rich loop that forms part of the ATP-binding pocket in porcine adenylate kinase (PAK), and site 5 corresponded to a region in PAK, located on one lobe of the cleft, that contains arginine residues that bind s...
متن کاملTwo forms of thymidine kinase in normal and tumor tissues of animals.
The thymidine kinases extracted from the spleen of mice infected with Friend virus and from Yoshida sarcoma in rats were separated into two active peaks by diethylaminoethyl cellulose column chromatography, while those of normal tissues have been found to consist of only the first peak (P-1). The second peak (P-II) was also found in the enzyme from the extract of the spleen when the animals wer...
متن کاملHuman deoxythymidine kinase. I. Purification and general properties of the cytoplasmic and mitochondrial isozymes derived from blast cells of acute myelocytic leukemia.
Two forms of deoxythymidine kinase from blast cells of acute myelocytic leukemia were identified by electrophoresis. One was associated mainly with the cytoplasm and the other with mitochondria. Both isozymes were separated and purified by differential affinity column chromatography which resulted in 2416- and 1634-fold purification of the cytoplasmic and mitochondrial enzymes, respectively. Af...
متن کاملIntegrin Linked Kinase (X-ILK) Function during Embryonic Development and within Adult Tissues of Xenopus laevis
Integrin linked kinase (ILK) is a serine/threonine protein kinase implicated in the phosphatidylinositol 3’kinase (PI3’K) pathway. Integrin linked kinase has been investigated in different organisms such as mammalian systems (human, mice, rat), insects (Drosophila) and nematodes (Cenorhabditis elegans), however to date little data regarding ILK research on amphibians has been reported. In...
